Abstract

Objective It is to explore the protective effect of calcitriol on cerebral ischemia-reperfusion injury in rats and its effect on mitochondrial autophagy.Methods Thirty-two SD rats were randomly divided into sham operation group,model group,normal saline group and osteotriol group,with 8 rats in each group.The osteotriol group was given osteotriol 2 μg/kg by gavage,and the model group and normal saline group were given normal saline 2 mL/kg by gavage,all once daily,continuously treated for 7 days.On the 8th day of the experiment,the rats of all groups except for the sham operation group were prepared to establish models of cerebral ischemia/reperfusion injury by wire suturing.The neurological deficits of the rats were evaluated by Longa's method,the cerebral infarction was observed by TTC staining,and the volume of cer-ebral infarction was calculated,the mitochondrial membrane potential was detected by JC-1 staining,the mitochondria ul-trastructure of cerebral cortex on the injured side was observed by transmission electron microscopy,and the expressions of mitochondrial function-related proteins NIP3-like protein X(NIX),microtubule-associated protein 1(Beclin1),dynamin-related protein 1(DRP1),optic atrophy 1(OPA1)and Peroxisome proliferator activated receptor gamma coactivator-1α(PGC-1α)in cerebral cortex on the injured side were measured by Western blotting.Results Compared with the sham op-eration group,the neurological deficit scores and cerebral infarction volumes of rats in the model group and normal saline group were significantly increased(all P<0.05);the red/green fluorescence intensity ratios in the mitochondrial mem-brane potential detection were significantly decreased(both P<0.05);the mitochondrial damage in the cerebral cortex was severe;the relative protein expressions of NIX,Beclin1 and DRP1 in cerebral cortex on the injured side were signifi-cantly increased(all P<0.05),while the relative protein expressions of OPA1 and PGC-1α were significantly reduced(all P<0.05).Compared with the model group and normal saline group,the neurological deficit scores and cerebral in-farction volume of rats in the osteotriol group were significantly reduced(all P<0.05);the red/green fluorescence intensi-ty ratios in the mitochondrial membrane potential decoction were significantly increased(both P<0.05);the mitochondrial damage in the cerebral cortex was obviously alleviated;the relative protein expressions of NIX,Beclin1 and DRP1 in cere-bral cortex on the injured side were significantly reduced(all P<0.05),and the relative protein expressions of OPA1 and PGC-1α were significantly increased(all P<0.05).Conclusion Osteotriol can protect brains of rats with cerebral ische-mia-reperfusion injury,which may maintain mitochondrial ultrastructure and thus protect neurons by modulating mitochon-drial autophagy,attenuating mitochondrial energy depletion,increasing mitochondrial biosynthesis and other processes.
