Abstract

To gain a deeper understanding of the molecular mechanisms of secondary metabolism in citrus and to enhance the nutritional value of citrus through metabolic engineering,this study focused on sweet oranges and employed bioinformatics methods to identify and analyze CsMYC transcription factors across the entire genome.The research identified a total of 14 CsMYC transcription factor members,dis-tributed across four chromosomes of sweet oranges.Phylogenetic analysis indicated that the CsMYC tran-scription factor family in sweet oranges could be classified into four subfamilies,and most genes within the same subfamily shared similar gene structures and conserved motifs.Analysis of transcriptome data from Fengjie '72-1' sweet oranges and Cleopatra sweet oranges revealed that CsMYC1 was highly ex-pressed in the juice sacs of young fruits,CsMYC5 was highly expressed in various tissues of young fruits,and CsMYC11 was highly expressed in the albedo(AL)and segment membranes(SM)of young fruits.Additionally,CsMYC1,CsMYC2,CsMYC5 and CsMYC11 were highly expressed under heat stress(HS),water stress,and combined heat and water stress(WS+HS).Protein interaction analysis showed that the Arabidopsis homologous genes AtMYC1,AtBHLH13,AtBHLH3,and AtMYC2 of CsMYC1,CsMYC2,CsMYC5 and CsMYC11(respectively)directly or indirectly regulated flavonoid synthesis.These four genes were involved in plant defense and might all be involved in the biosynthesis of flavonoids in citrus fruits.The research results laid the foundation for the functional study of CsMYC genes.
