绵羊肺炎支原体毒力评价细胞模型的建立
Establishment of a cell model for evaluating the virulence of Mycoplasma ovipneumoniae

Abstract
To establish a cell model for virulence evaluation of the Mycoplasma ovipneumoniae by detecting and analyzing the virulence-related factors of cells infected with M.ovipneumoniae.In this study,5 M.ovipneumoniae strains with known virulence of 108 CCU50/mL were employed to infect the MDBK cell.IFA and quantitative PCR method were used to detect the adhesion ability of M.ovipneumoniae strains.Cytopathogenic effect(CPE)was observed.And cell viability,cytotoxicity,and intracellular H2O2 content were measured by using the commercial kits.According to the results of significance analysis,the virulence of 5 M.ovipneumoniae strains was evaluated.Base on the results of correlation analysis,the most relevant virulence evaluation factors of M.ovipneumoniae strain were screened.The adhesion ability,damage effect on cell activity,damage effect on cell membrane integrity,and mediating effect of IK3-3 and NJ01 were stronger than those of Y98,N3-2F and XLY.The results indicated that IK3-3 and NJ01 were highly virulent strains,N3-2F and XLY were moderately virulent strains,and Y98 was a weak virulence strain,which was consistent with the results of the previous animal infection test.The results of correlation analysis showed that the virulence evaluation factors of the cell infection virulence evaluation model were feasible,and the adhesion ability of M.ovipneumoniae was the most correlated factor with its virulence.The cell model established in this study provides a new test platform for evaluating virulence,studying the pathogenic mechanism and screening vaccine candidates of M.ovipneumoniae.关键词
