摘要

Abstract

To investigate the inhibitory effect of the antioxidant derivative Q-Griffithsin(Q-GRFT)on porcine epidemic diarrhea virus(PEDV),a recombinant expression plasmid pGEX-4T-Q-GRFT was constructed,and the recombinant protein Q-GRFT was expressed using a prokaryotic expression system.The target protein was purified by nickel-affinity chromatography and its specificity of the concen-trated protein was validated.Its anti-PEDV effect was further confirmed by in vitro experiments.The results showed that the pGEX-4T-Q-GRFT was successfully transformed into competent cells.Following induction at 22 ℃ for 14 h,the Q-GRFT protein was expressed at its anticipated molecular weight,with a purification yield of 3.01 mg/L.In vitro assay,Q-GRFT markedly mitigated the cytopathic effects induced by PEDV infection,inhibited PEDV replication within cells,and demonstrated antiviral activity in a mass concentration-dependent.In summary,this study successfully prepared a recombinant protein Q-GRFT possessing anti-PEDV activity,providing a theoretical foundation for the development of antiviral agents against animal coronaviruses,and proposing innovative strategies for PEDV prophylaxis and treatment.
