Microbial intervention can alleviate cadmium-induced liver and kidney function damage and oxidative stress in rats

摘要

Abstract

In order to investigate the effect of microbial intervention on cadmium-induced liver and kidney function damage in rats.Fifty SPF female Wistar rats were randomly divided into five groups after one week of adaptive feeding,namely,blank control group(Con),probiotic control group(MP),CdCl2 treatment group(Cd),probiotic intervention group(Cd+MP),and fecal microbiota transplantation inter-vention group(Cd+FMT).Rats in Cd group,probiotic intervention group,and fecal microbiota transplan-tation intervention group were gavaged with cadmium chloride(CdCl2)at a dose of 10 mg per kilogram of body weight per day,and rats in Con group and MP group were gavaged with equal volumes of double-distilled water.After 4 weeks of treatment,rats in the MP group and probiotic intervention group were gavaged with probiotics at a dose of 5× 108 CFU per day.Rats in the fecal microbiota transplantation interven-tion group were gavaged with fecal bacterial suspension from the rats of Con group at a dose of 10 mL per kilogram body weight per day.Rats in the Con group and the Cd group were gavaged with equal volumes of double-distilled water.After 4 weeks,the body weight of rats was recorded.The rats were anesthetized,sacrificed by cervical dislocation,blood was collected from the heart,and liver,kidney tissues were dissected and weighed.The activities of alkaline phosphatase(ALP),alanine aminotransferase(ALT)and aspartate aminotransferase(AST),and the contents of creatinine(CREA)and urea(UREA)in serum,the ac-tivities of catalase(CAT)and total superoxide dismutase(T-SOD),the contents of malondialdehyde(MDA)and reduced glutathione(GSH),and total antioxidant capacity(T-AOC)in liver and kidney tis-sues were determined by colorimetry.The results showed that probiotics intervention and fecal micro-biota transplantation intervention significantly or extremely significantly inhibited cadmium-in-duced weight loss,the increase of liver and kidney index,and the increase of ALP,ALT,AST activities and CREA content in serum,the increase of CAT,T-SOD activities,MDA content and T-AOC,and the decrease of GSH content in liver and kidney of rats(P<0.05 or P<0.01).The above results indicate that microbial intervention can effectively alleviate cadmium-induced liver and kidney function damage and oxidative stress in rats.
