鸭疫里氏杆菌不同临床分离株的自然转化频率和耐药谱鉴定与分析
Identification and Analysis of Natural Transformation Frequency and Antibiotic Resistance Profile among Different Clinical Isolates of Riemerella anatipestifer

Abstract
Riemerella anatipestifer(RA)can infect various poultry species,and some clinical isolates exhibit multiple drug resistances,with resistance genes being able to spread between bacteria through natural transformation.However,it remains unclear whether there are differences in the natural transformation frequency among different RA clinical isolates,and whether the growth characteristics and resistance profiles of the strains are related to their natural transformation frequencies.In this study,42 RA clinical isolates were tested for their natural transformation frequency using the tonB1up-cfxA-tonB1down gene fragment(DNA fragment carrying the resistance gene)as the substrate DNA.The OD600 nm values of bacterial cultures at different growth times were measured to plot the growth curve.The minimum inhibitory concentration(MIC)of 10 antibiotics from 5 major categories were determined for the 42 RA clinical isolates,and the relationship between natural transformation frequency,growth characteristics,and resistance profiles was analyzed.The results showed that the natural transformation frequency among the 42 RA clinical isolates ranged from a maximum of(9.78±3.6)×10⁻⁵ to below the detection limit(<10⁻⁹).The growth curve analysis revealed that the highest OD600 nm value at 14 hours of growth for the RA clinical isolates was 5.2±0.3,while the lowest was 3.1±0.5.Among the RA clinical isolates,88.10%(37/42)exhibited multiple drug resistances,with the highest resistance to 7 drugs.23.81%(10/42)of the isolates were resistant to more than 5 drugs,and 40.48%(17/42)were resistant to more than 4 drugs.Further analysis showed that the natural transformation frequency was not significantly correlated with growth characteristics or the resistance profiles(the Pearson correlation coefficient was less than 0.8 in all cases).This study provides data support for exploring the relationship between the increase in RA's resistance profile and its natural transformation frequency.关键词
