Effect of Glucoronolactone on Intestinal Health in Weaned Piglets Challenged with PRRSV and Deoxynivalenol

Abstract
To investigate the effect of glucuronolactone(GLU)on intestinal health in piglets jointly challenged with porcine reproductive and respiratory syndrome virus(PRRSV)and deoxynivalenol(DON),30 weaned piglets(21 days old)were randomly divided into three groups:Control group(fed a basal diet),DON group(fed a basal diet containing 2 mg/kg DON),and DON+GLU group(fed a basal diet containing 2 mg/kg DON and 200 mg/kg GLU).All piglets were fed their respective diets from day 1,and on day 15,each piglet was intramuscularly injected with 2.0 mL of PRRSV(10⁵ TCID₅₀/mL).Fecal scores were recorded daily post-challenge to calculate the average diarrhea rate.After 3 weeks of challenge,all piglets were sampled for further analyses.Nutrient digestibility was assessed using an indicator method.Jejunal and ileal morphological structures were examined via hematoxylin-eosin(H.E.)staining,followed by pathological scoring.Western blot analysis was used to detect the expression of intestinal mucosal proteins zonula occludens-1(ZO-1),Occludin,and Claudin-1,while real-time fluorescence quantitative PCR(qPCR)was performed to determine the mRNA expression of interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-8(IL-8),tumor necrosis factor alpha(TNF-α),ZO-1,Occludin,and Claudin-1 in jejunal and ileal mucosa.Additionally,fecal microbial diversity was analyzed via 16S rDNA high-throughput sequencing.The results showed that compared with the Control group,PRRSV-challenged piglets in the DON group showed a significantly higher average diarrhea rate(P<0.05),reduced apparent digestibility of crude fat and energy(P<0.05),and severe intestinal injury with significantly higher pathological scores in the jejunum and ileum(P<0.05).The villus height to crypt depth ratio(V/C)was significantly decreased(P<0.05).The mRNA expression levels of Occludin and Claudin-1 in the jejunum,as well as protein expression levels of ZO-1,Occludin in the jejunum,and ZO-1,Occludin and Claudin-1 in the ileum,were significantly reduced(P<0.05).Meanwhile,the mRNA expression of IL-1β,TNF-α(jejunum),and IL-8,TNF-α(ileum)were significantly increased(P<0.05).The microbial diversity indices(Chao1,Shannon,Simpson)were all significantly decreased(P<0.05).Compared with the DON group,the DON+GLU group showed a trend toward reduced diarrhea rate(P>0.05),a significant increase in apparent digestibility of crude fat and energy(P<0.05),and milder intestinal injury with lower pathological scores and higher V/C ratio(P<0.05).The mRNA expression of ZO-1 and Occludin in both jejunum and ileum was significantly increased(P<0.05),along with increased Occludin protein expression(P<0.05).The mRNA expression levels of IL-8,TNF-α(jejunum)and IL-1β,IL-6,IL-8(ileum)were significantly decreased(P<0.05).Microbial diversity indices(Chao1,Shannon,Simpson)were significantly elevated(P<0.05).In conclusion,dietary supplementation with GLU alleviated PRRSV-DON co-induced intestinal barrier damage,inflammation,and gut microbial dysbiosis in weaned piglets.These findings suggest that GLU could serve as a novel nutritional intervention to counteract the synergistic effects of PRRSV and DON,providing a theoretical foundation for its application in piglet production.关键词
