Isolation and Identification of Bovine Enterovirus

Abstract
To investigate the prevalence of bovine enterovirus(BEV)in China and provide theoretical support for its prevention and control,a virus strain was isolated from the feces of diarrheic cattle in a farm in Chengdu,Sichuan Province.The strain was designated SC-726 and subjected to further study.After inoculation of SC-726 into Madin-Darby bovine kidney(MDBK)cells,cytopathic effects(CPE)were observed,and viral titers were calculated.Transmission electron microscopy(TEM)was used to observe the viral morphology,and its physicochemical properties,nucleic acid type,and cell tropism were analyzed.A one-step growth curve was constructed,and the 5' untranslated region(5'UTR)of the viral genome was sequenced for phylogenetic analysis.The results showed that SC-726 caused significant CPE in MDBK cells,with a peak titer of 1×106.2 TCID₅₀/0.1 mL.TEM revealed non-enveloped spherical particles approximately 30 nm in diameter,consistent with the morphology of typical picornaviruses.Physicochemical analysis indicated that the isolate was resistant to organic solvents(ether and chloroform)and trypsin,and showed properties characteristic of BEV:acid resistance,alkali sensitivity,and heat lability.The DNA synthesis inhibitor Ara-C had no effect on viral replication,confirming that SC-726 is an RNA virus.The strain could replicate in multiple animal cell lines,including MDBK,baby hamster Syrian kidney(BHK-21),porcine kidney-15(PK-15),African green monkey embryonic kidney(Marc-145),and Madin-Darby canine kidney(MDCK)cells.Phylogenetic analysis based on the 5'UTR sequence classified SC-726 as a BEV-F subtype.In conclusion,one BEV-F strain was successfully isolated from diarrheic cattle feces,contributing to the genetic database of BEV in China and providing a theoretical basis for further prevention and control strategies.关键词
