基于DIA技术的多药耐药性捻转血矛线虫蛋白质组学分析
Proteomic Analysis of Multidrug Resistance of Haemonchus contortus by DIA Technology

Abstract
To explore the mechanisms underlying the development of anthelmintic resistance,this study treated Haemonchus contortus third stage larvae(L3)with ivermectin,albendazole,and levamisole,and employed data-independent acquisition(DIA)proteomic sequencing to analyze the differential protein expression before and after drug treatment,to identify multidrug resistance-related proteins.The results revealed a total of 1 888 differentially expressed proteins,of which 63 were related to multidrug resistance.Further analysis revealed 36 proteins with significantly different expression(P<0.05),of which 25 were specific to Haemonchus contortus and related to drug resistance.Gene ontology(GO)analysis classified 60 of these differential proteins into 76 biological process branches,while Kyoto encyclopedia of genes and genomes(KEGG)analysis revealed that 46 differentially expressed proteins were enriched in 75 signaling pathways related to lipid metabolism and endocrine systems.Comprehensive analysis identified five Haemonchus contortus specific drug resistance proteins with significant expression differences:proteins containing UDP-glucuronosyltransferase domains,antigen H11,immunoglobulin v and i groups,proteins with protein tyrosine kinase(PTK)domains,and proteins with the lethal giant larvae homolog 2 domain.These proteins are involved in important biological processes and key metabolic pathways.The results of this study provide foundational data for understanding the relationship between differentially expressed proteins and lipid metabolism,endocrine systems,and multidrug resistance in Haemonchus contortus,and have significant implications for revealing the mechanisms underlying multidrug resistance in this parasitic nematode.关键词
