马齿苋发酵液对小鼠细菌性腹泻的防治效果评价
Evaluation of the Preventive and Therapeutic Effect of Portulaca oleracea Fermentation Liquid on Bacterial Diarrhea in Mice

Abstract
To evaluate the preventive and therapeutic effect of Portulaca oleracea fermentation liquid on diarrhea induced by pathogenic Escherichia coli(E.coli)infection in mice,this study selected 30 ICR mice,randomly divided into 6 groups,with 5 mice per group.Each group was orally administered 0.02 mL/(g·bw)of the respective solution.The Blank group and Model group were given distilled water,the Positive control group was given Enteritis Ning tablets(32.5 mg/mL),and the Low-,Medium-,and High-dose of Portulaca oleracea fermentation liquid groups were administered at concentrations of 1.2,2.4,and 4.8 g/mL,respectively,for 7 consecutive days.Two hours after the final dose,except for the Blank group which received saline intraperitoneally,all other groups were intraperitoneally injected with pathogenic E.coli to establish the mouse diarrhea model.Diarrhea incidence and diarrhea index were observed,small intestine morphology was assessed,and the contents of intestinal mucin 2(MUC2),serum interleukin-6(IL-6),interleukin-12(IL-12),interleukin-2(IL-2),tumor necrosis factor-alpha(TNF-α),and chemokine ligand 3(CCL3)were measured.The results showed that the Model group had the highest diarrhea rate and diarrhea index,with the most severe damage to the intestinal villi.Compared with the Blank group,the Model group had significantly reduced MUC2 expression in the intestine(P<0.05),and significantly elevated serum levels of IL-6,IL-12,IL-2,TNF-α,and CCL3(P<0.05).Compared with the Model group,Portulaca oleracea fermentation liquid reduced the diarrhea rate and diarrhea index in mice,effectively improved the damage to the intestinal villi,and significantly increased MUC2 expression in the intestinal tissues of the Medium-and High-dose groups(P<0.05),Furthermore,serum IL-6,TNF-α,and CCL3 levels were significantly reduced in the Low-,Medium-and High-dose groups(P<0.05),and IL-2 levels were significantly reduced in the Low-and Medium-dose groups(P<0.05).These results suggest that Portulaca oleracea fermentation liquid can reduce the incidence of E.coli-induced diarrhea in mice,improve the morphology of the small intestine,alleviate inflammatory responses,and provide protective effects against bacterial diarrhea in animals.关键词
