摘要

Abstract

With the progress of organ transplantation technology,more and more patients with end-stage diseases have gained new life.In order to meet the global demand for organs,xenotransplantation technology has been developed rapidly in recent years.Transplantation of non-human primate(NHP)derived kidneys,hearts and livers has been developed,with increasing number of trials using pig-derived organs.The gene editing technology based on clustered regularly interspaced short palindromic repeats(CRISPR)can achieve the modification of multiple gene loci.Researchers are developing gene-edited pig lines by altering the response of natural antibodies to the vascular endothelium of porcine organs,activating the complement system and intrinsic immune cells,and inducing coagulation cascade,inflammatory and adaptive immune responses.The first human trial of xenogeneic heart transplantation was carried out in early 2022.The technology of xenotransplantation has gradually entered the clinical practice,but it is also facing many challenges in medicine,ethics and supervision.Researchers should be more cautious in evaluating the quality of medical products,and all parties should strengthen communication and exchange to promote the development of global transplantation.This paper summarizes the technological development route in this field,and ponders over the future development trend and challenges.
