Circulating bile acids influence the risk factors of cardiovascular diseases in patients with polycystic ovary syndrome

摘要

Abstract

Objective To investigate the alterations of bile acids in overweight/obese polycystic ovary syndrome(PCOS)after the intervention with calorie-restricted diet(CRD)or dulaglutide combined with CRD and its association with cardiovascular disease risk factors.Methods This trial was a randomized,controlled,open-label study conducted in Shanghai Tenth People's Hospital between May 2021 and May 2022.A total of overweight/obese PCOS patients were randomly assigned to CRD alone(n=33)or dulaglutide combined with CRD(n=35)within 6 months.The serum bile acid profiles before and after CRD or dulaglutide combined with CRD treatment were detected by a developed liquid chromatography-tandem mass spectrometry(LC-MS/MS)method.Additionally,the correlation between different bile acid metabolites and risk factors for cardiovascular diseases in PCOS patients was explored.Results After the CRD treatment,lithocholic acid(LCA)and taurolithocholic acid(TLCA)significantly increased in the serums of overweight/obese PCOS patients.The combination of dulaglutide and CRD led to significant differences in the primary conjugated bile acids levels.Moreover,dulaglutide combined with CRD treatment dramatically increased circulating TLCA,cholic acid(CA),and LCA levels,while serum glycochenodeoxycholic acid(GCDCA)decreased.Correlation analysis indicated that changes of bile acid metabolites in overweight/obese PCOS patients after treatment were associated with the alterations of clinical characteristics which were related to cardiovascular diseases.Conclusions Our data suggested that CRD or the combination of dulaglutide and CRD altered bile acids metabolism in overweight/obese PCOS patients.Bile acids metabolism was associated with the improvement of clinical parameters such as risk factors for cardiovascular diseases.
