摘要

Abstract

Objective To investigate the expression of celluar senescence-related genes in microglia of mice with diabetic encephalopathy.Methods Single-cell sequencing technique was used to map the hippocampus of diabetic encephalopathy mice.The expression of cellular senescence related genes in microglia was detected by qRT-PCR.Results The expression levels of cellular senescence related genes Calm2,Mapkapk2,Nfkb1,H2-D1,Ccnd3,Cdk6 and Il-1α were determined by single-cell mapping and dimensional clustering analysis of the hippocampus in diabetic encephalopathy.qRT-PCR showed that the mRNA expression levels of Mapkapk2,Nfkb1,H2-D1,Cdk6 and Il-1α were significantly increased in BV2 cells treated with high glucose,while the mRNA expression levels of Calm2 and Ccnd3 were significantly decreased.Conclusions Single-cell sequencing revealed that diabetic encephalopathy affects gene expression associated to microglial senescence,providing a molecular mechanism and potential therapeutic target for diabetic encephalopathy.
