基于网络药理学与动物实验探讨经典名方栀子大黄汤治疗肝纤维化的作用机制
Investigation on the function mechanism of the classic formula Zhizi Dahuang Decoction in the treatment of liver fibrosis based on network pharmacology and animal experiments

Abstract
Objective To investigate the function mechanism of Zhizi Dahuang Decoction in the treatment of liver fibrosis(LF)based on network pharmacology and animal experiments.Methods A 0.1%percentage of 3,5-diethoxycarbonyl-1,4 dihydroclonidine(DDC)feed was used to establish an animal model of LF in mice,and the changes of serum indices in mice were observed to investigate the therapeutic efficacy of Zhizi Dahuang Decoction on LF.Obtaining LF-related targets from the database,they were intersected with components of Zhizi Dahuang Decoction.Protein-protein interaction analysis of intersection targets was carried out by using STRING database.Enrichment analysis of gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)was carried out.Molecular docking technology was used to verify the binding effect of core components and key targets.Results Zhizi Dahuang Decoction could alleviate LF induced by DDC feed.A total of 49 active components of Zhizi Dahuang Decoction were screened out,and 178 potential targets for treating LF were screened out.The enrichment analysis of GO and KEGG showed that Zhizi Dahuang Decoction was mainly used in the treatment of LF,through biological processes such as positive regulation of cell migration,it played the role of DNA-binding transcription factor binding,and played an anti-LF function by regulating signal pathways such as phosphatidylinositol 3-kinase-protein kinase B(PI3K-Akt).The results of molecular docking showed that both core components and core targets had relatively strong binding force.Conclusion It is preliminarily confirmed that Zhizi Dahuang Decoction has a significant therapeutic efficacy on LF mice,which may be caused by multiple compounds acting on multiple molecular targets.Its molecular mechanism is mainly to inhibit inflammatory reaction and regulate PI3K-Akt and other signal pathways.关键词
