Abstract

This study aimed to investigate the preventive effect and mechanism of asiatic acid on Salmonella-induced colitis,providing a basis for the development of new veterinary drugs from Chinese medicine.24 BALB/c mice were randomly divided into a control group,a Salmonella infection group and an asiatic acid intervention group.The treatment was given for 7 days,with the control and Salmonella infection groups receiving normal saline,while the asiatic acid intervention group was administered with asiatic acid at a concentration of 0.4 mg/mL and a dose of 10 mg/kg.On the 8th day,the control group continued to receive normal saline,while the Salmonella infection group and the asiatic acid intervention group were challenged with 1×109 cfu of Salmonella typhimurium.Observations were made on the effects of asiatic acid intervention on colon damage,inflammatory cytokine expression,intestinal mucosal barrier function and gene expression changes induced by Salmonella infection in mice.H-E staining results showed that asiatic acid could effectively alleviate the shortening of crypts,inflammatory cell infiltration and congestion in the colon of mice induced by Salmonella.PAS staining indicated that asiatic acid could mitigate the reduction in the number of colonic goblet cells caused by Salmonella.Quantitative PCR results demonstrated that asiatic acid significantly inhibited the increase in IL-6,IL-1β,and TNF-α mRNA expression levels induced by Salmonella(P<0.05).Western blot analysis revealed that asiatic acid effectively prevented the downregulation of tight junction proteins claudin-2 and claudin-7 caused by Salmonella(P<0.05).Transcriptome sequencing results suggested that asiatic acid could partially reverse the abnormal gene expression caused by Salmonella,which was associated with inflammation and ion transmembrane transport pathways.The results showed that asiatic acid could alleviate Salmonella-induced colitis,indicating its potential as a pharmacologically active ingredient for the prevention and treatment of Salmonella infection.
