Abstract

This study aimed to investigate the inhibitory effect of short-tailed phage P6 on multi-drug resistant Salmonella,so as to provide experimental basis for the development of novel antimicrobial strategies.In this experiment,the inhibitory effects of P6 on 43 multi-drug resistant isolates of S.typhimurium and S.enteritidis in floating state and biofilm state were systematically evaluated by double-agar plate method and 96-well plate crystal violet staining method.The results showed that after using the double-layer agar plate method to inhibit 43 strains of multidrug-resistant Salmonella with phage P6,phage P6 could lyse 51.2%of the floating drug-resistant Salmonella strains,and the lysate rate of S.enteritidis was 40.0%(4/10)and that of S.typhimurium was 54.5%(18/33).The most common resistance profile among the 43 Salmonella strains was PG-AMX-AMP-STR(44.2%),and the lytic activity of phage P6 against Salmonella strains with this resistance profile reached 93.8%(15/16).At the same time,the lytic phage P6 could inhibit the biofilm of different multi-drug resistant Salmonella by degrading extracellular polymeric substances,and the phage P6 could reduce the biofilm by 11.2%-88.6%within 3 hours.The highest biofilm reduction was achieved in 78.4%of resistant S.enteritidis and 88.6%of resistant S.typhimurium.The average biofilm reduction rate of P6 against resistant S.typhimurium was 45.2%,and the average biofilm reduction rate against resistant S.enteritidis was 64.4%.This experiment proved that P6 had certain inhibitory effects on the floating and biofilm states of the two serotypes of multidrug-resistant Salmonella,which could provide important data support for the optimization and promotion of phage therapy,and also lay the foundation for the combined application of phage with other antibacterial agents.
