不同水位育苗方式对烤烟幼苗根系和耐冷性的影响OA
Effects of Different Seedling Raising Methods at Different Water Levels on Root System and Cold Tolerance of Tobacco Seedlings
为探索不同水位育苗方式对烤烟幼苗根系和耐冷性的影响,以烤烟品种湘烟 7 号为材料,设置浅水位和深水位 2 种育苗方式处理,对不同处理烤烟的种子萌发情况、幼苗根系表型、不同光照条件(0、4 000 Lux)下遭受低温胁迫(4℃)后的生理活性等指标进行比较分析.结果表明:与深水位育苗处理相比,浅水位育苗处理烤烟的发芽率、发芽势分别提高 6.25%和 21.88%,根长、根表面积、根尖数和根分叉数分别提高 1.06 倍、16.76%、81.51%和 43.42%,低温处理 0、6、12 h 后的根系活力分别提高 12.64%、44.47%和 51.87%.无论在适宜光照(4 000 Lux)还是无光条件(0 Lux)的冷胁迫下,浅水位育苗处理的过氧化物酶(POD)、过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性显著高于深水位育苗处理,而且 2 个处理均为适宜光照条件下的抗氧化酶活性水平更高.低温胁迫前后不同育苗处理的差异表达基因富集分析结果显示,浅水位育苗处理的能量转化能力较强,应对冷胁迫时可产生较多的应对胁迫的产物(如萜烯合酶和肽酶).综合分析,浅水位育苗处理有利于培养较发达的深层根系,增强根系活力,提高抗氧化酶活性,可改善烤烟幼苗应对冷胁迫的能力.
In order to explore the effects of different water level seedling cultivation methods on the root system and cold tolerance of flue-cured tobacco seedlings,flue-cured tobacco variety Xiangyan No.7 was used as material,and two seed-ling cultivation methods were set up in shallow water level and deep water level.The seed germination,root phenotype,physiological activity and other indicators of flue-cured tobacco seedlings under different light conditions(0,4 000 Lux)were compared and analyzed.The results showed that compared with the deep water seedling treatment,the germination rate and germination energy of flue-cured tobacco seedlings treated in shallow water seedling treatment increased by 6.25%and 21.88%,respectively,and the root length,root surface area,root tip number and root bifurcation number increased by 1.06 times,16.84%,81.5%and 43.42%,respectively.The root activity after 0,6,12 hours of low temperature treatment increased by12.64%,44.47%and51.87%respectively.Whether under cold stress under suitable light(4 000 Lux)or no light(0 Lux)conditions,the activities of POD,CAT and SOD in shallow water seedling treatment were significantly higher than those in deep water seedling treatment,and both treatments had higher levels of antioxidant enzyme activities under suitable light conditions.The enrichment analysis results of differentially expressed genes in different seedling treat-ments before and after low temperature stress showed that the seedling treatment in shallow water had strong energy conver-sion ability,and more products that responded to cold stress(such as terpene synthase and peptidases)could be produced when responding to cold stress.Comprehensive analysis shows that the shallow water seedling treatment is conducive to cul-tivating more developed deep root systems,enhancing root vitality,increasing antioxidant enzyme activities,and improving the ability of flue-cured tobacco seedlings to cope with cold stress.
