基于孟德尔随机化分析的薏苡附子散治疗急性心肌缺血的分子机制研究
Molecular Mechanism of Yiyi Fuzi Powder in Treating Acute Myocardial Ischemia Based on Mendelian Randomization Analysis

Abstract
Objective:This study aimed to investigate the potential molecular mechanisms of Yiyi Fuzi Powder in the treatment of acute myocardial ischemia(AMI)by integrating Mendelian randomization(MR)analysis,thereby providing new approaches for the fundamental research of Yiyi Fuzi Powder applied in the treatment of cardiovascular diseases.Methods:First,network pharmacology was employed to identify the main chemical constituents and targets of Yiyi Fuzi Powder.Subsequently,MR analysis was conducted to evaluate the causal relationship between these targets and AMI.Finally,molecular docking was utilized to simulate the interactions between the active ingredients of Yiyi Fuzi Powder and key target proteins,assessing their binding modes and stability.Results:The bioactive compounds in Yiyi Fuzi Powder,namely[(2R)-2,3-dihydroxypropyl](Z)-octadec-9-enoate,2-monoolein,delphinidin,carnosifloside,isotalatizidine,deoxyandrographolide,deltoin,karanjin,and benzoylnapelline,have the potential to modulate the expression of six key protein targets,which are death-associated protein kinase 1(DAPK1),fatty acid amide hydrolase(FAAH),glutathione S-transferase mu 2(GSTM2),MAP kinase-interacting serine/threonine-protein kinase 1(MKNK1),phosphodiesterase 8B(PDE8B),and prostaglandin E receptor 4(PTGER4),the genes encoding these targets were found to have significant causal relationships with AMI.Among these targets,DAPK1 and PDE8B are negatively correlated with AMI,while FAAH,GSTM2,MKNK1,and PTGER4 are positively correlated with AMI.Molecular docking analysis further reveals that the active components of Yiyi Fuzi Powder exhibit stable binding interactions with these critical targets.Conclusion:The study further predicts the key components and targets of Yiyi Fuzi Powder in the treatment of AMI using network pharmacology,MR analysis,and molecular docking techniques.The results suggest that Yiyi Fuzi Powder may exert its therapeutic effects by upregulating the expression of DAPK1 and PDE8B while inhibiting the expression of FAAH,GSTM2,MKNK1,and PTGER4.These actions may promote anti-inflammatory,anti-apoptotic,and cell stress response-inhibitory effects,thus achieving the goal of treating AMI.This provides a new perspective for the clinical research of Yiyi Fuzi Powder.关键词
