Identification of Saposhnikoviae Radix and Its Adulterants and Counterfeits by UPLC-Q-TOF-MS and Microscopic Characterization

Abstract
Objective:To establish the identification methods for distinguishing Saposhnikovia divaricata(Turcz.)Schischk.and its adulterants and counterfeits.Methods:Morphological and microscopic analyses were performed for preliminary differentiation between S.divaricata and its adulterants(Libanotis laticalycina Shan et Shen)or substitutes(Peucedanum praeruptorum Dunn,Seseli mairei Wolff,and Pimpinella candolleana Wight et Arn.).The contents of prim-O-glucosylcimifugin and 5-O-methylvisamminoside in the samples were determined using high-performance liquid chromatography(HPLC).Ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS)was employed to analyze the main components of S.divaricata and its adulterants and counterfeits.The orthogonal partial least squares-discriminant analysis(OPLS-DA)was adopted to compare the primary and secondary metabolites between S.divaricata and its adulterants and counterfeits.The Kyoto Encyclopedia of Genes and Genomes(KEGG)was employed to enrich the metabolic pathways.Results:Differences were observed in appearance and microscopic structures between Saposhnikoviae Radix and its adulterants and counterfeits.Measurement of total prim-O-glucosylcimifugin and 5-O-methylvisamminoside content effectively distinguished S.divaricata from substitutes.Based on the OPLS-DA method,differential secondary metabolites such as coumarins and flavonoids were screened by setting fold change(FC)and frequency error,followed by enrichment into secondary metabolic pathways associated with these differential metabolites.Conclusion:S.divaricata can be authenticated through integrative approaches including morphological evaluation,microscopic examination,quantitative metabolite analysis,and metabolomics profiling.关键词
