基于PI3K/Akt和MAPK信号通路探讨茶芎对偏头痛的作用机制
Mechanism of Action of Chaxiong in Treating Migraine Based on PI3K/Akt and MAPK Signaling Pathways

Abstract
Objective:To explore the mechanism of action of Chaxiong in treating migraine based on PI3K/Akt and MAPK signaling pathways.Methods:Thirty-six male SD rats were randomly divided into six groups:a control group,a model group,groups receiving low-,medium-,and high-doses of Chaxiong(CX-L,CX-M,and CX-H groups),and a group receiving flunarizine hydrochloride(FNZ group),with six rats in each group.Each group was given a corresponding drug or normal saline intragastric administration for five consecutive days.Half an hour after the last administration,except for the control group,all groups were given subcutaneous injections of nitroglycerin(10 mg·kg-1)to induce a migraine model.Meanwhile,the control group was administered an equivalent volume of normal saline through subcutaneous injection.After modeling,the number of head scratching in rats within 0-60 minutes was recorded using the segmental counting method.The serum levels of endothelin(ET)and nitric oxide(NO),as well as brain tissue levels of tumor necrosis factor-α(TNF-α)and 5-hydroxytryptamine(5-HT),were determined using enzyme-linked immunosorbent assay(ELISA).Hematoxylin-eosin(HE)was used to observe the pathological changes in brain tissue,while Nissl staining was used to observe the number of Nissl bodies in brain tissue.Western blot and real-time fluorescent quantitative polymerase chain reaction(real-time PCR)were employed to detect the expression levels of PI3K/Akt and MAPK-related proteins and mRNAs in migraine rats.Results:Compared with the control group,the model group showed a significantly increased number of head scratching(P<0.001),decreased levels of serum ET and brain tissue 5-HT,as well as increased NO and TNF-α levels(P<0.05).The brain vessels of rats showed significant dilation and infiltration of inflammatory cells.Neuronal cell bodies displayed fewer Nissl bodies,and the proportion of positive cells decreased.The protein expression of p-PI3K,p-Akt,p-ERK,p-P38,and p-JNK was upregulated in brain tissue(P<0.05),and the gene expression of PI3K,Akt,ERK,P38,and JNK was significantly upregulated(P<0.05).Compared with the model group,a reduced number of head scratching(P<0.05),increased levels of ET and 5-HT,and decreased levels of NO and TNF-α(P<0.05 or P<0.01)were observed in the CX-L,CX-M,and CX-H groups.The degree of vascular dilation and the infiltration of inflammatory cells were reduced,while the number of Nissl bodies and the proportion of positive cells were increased.Additionally,Chaxiong treatment significantly downregulated the expression of p-PI3K,p-Akt,p-ERK,p-P38,and p-JNK proteins in brain tissue(P<0.05 or P<0.01),as well as the expression of PI3K,Akt,ERK,P38,and JNK genes(P<0.05).Conclusion:Chaxiong can significantly improve migraine in rats,and its mechanism is related to the regulation of PI3K/Akt and MAPK signaling pathways.关键词
