远志寡糖酯改善D-半乳糖致小鼠学习记忆障碍的作用
Effect of Oligosaccharide Esters from Polygala tenuifolia Willd.in Ameliorating D-galactose-induced Learning and Memory Impairment in Mice

Abstract
Objective:To investigate the effect of oligosaccharide esters from Polygala tenuifolia Willd.(OE)on learning and memory impairment in a rapidly-aging dementia mouse model induced by D-galactose.Methods:Eight-week-old Kunming mice were randomly divided into the control group,model group,donepezil group,and OE treatment groups(28.67,57.33 mg·kg-1).Brain aging was induced by intraperitoneal injection of D-galactose,and OE or donepezil was administered orally once daily for 60 consecutive days.Learning and memory abilities were assessed using the step-down test and the Morris water maze test.Serum was collected to measure superoxide dismutase(SOD)activity and malondialdehyde(MDA)levels.Immunohistochemistry was used to detect brain-derived neurotrophic factor(BDNF)expression in the cerebral cortex,while Western blot was performed to analyze the protein levels of protein kinase B(Akt),cAMP response element-binding protein(CREB),and BDNF in the cerebral cortex.Results:Compared with the control group,the model group exhibited significantly prolonged escape latency(P<0.05)and first platform-crossing latency(P<0.01)in the Morris water maze test,along with a significant reduction in platform-crossing frequency(P<0.01).In the step-down test,the first-jump latency was significantly shortened(P<0.01),while the number of errors increased significantly(P<0.01).Additionally,the model group showed significantly decreased serum SOD activity(P<0.01)and increased MDA levels(P<0.01),as well as significantly reduced levels of p-Akt/Akt,p-CREB/CREB,and BDNF in the cerebral cortex(P<0.01).Compared with the model group,OE treatment significantly shortened escape latency(P<0.05)and first platform-crossing latency(P<0.01),while increasing platform-crossing frequency(P<0.01)in the Morris water maze test.In the step-down test,OE prolonged the first-jump latency(P<0.01)and reduced the number of errors(P<0.01).Furthermore,OE enhanced serum SOD activity(P<0.05,P<0.01),reduced MDA levels(P<0.01),and upregulated the expression of p-Akt/Akt,p-CREB/CREB,and BDNF in the cerebral cortex(P<0.05,P<0.01).Conclusion:OE can alleviate learning and memory impairments in D-galactose-induced rapidly aging dementia mice,potentially by reducing oxidative damage and regulating the Akt/CREB/BDNF signaling pathway.关键词
