UPLC fingerprint combined with chemical pattern recognition to evaluate quality of Polygonatum cyrtonema Hua from different production areas

Abstract
Objective:To establish the fingerprint of Polygonatum cyrtonema Hua,and to evaluate the quality of Polygonatum cyrtonema Hua from different production areas by combining chemical pattern recognition.Methods:Ultra performance liquid chromatography(UPLC)was used for analysis with Waters HSS T3 C18 column(150 mm×2.1 mm,1.8 μm).The mobile phase was acetonitrile-0.1%phosphate water solution in gradient elution with the flow rate of 0.3 mL per minute.The column temperature was set at 30℃.Sample size was 1 μL and wavelength was 200 nm.The fingerprint of Polygonatum cyrtonema Hua was established,and analyzed by similarity,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA).The significant difference components were screened with variable importance projection index(VIP)value greater than 1.Results:The fingerprint of Polygonatum cyrtonema Hua identified 17 common peaks,and identified 2 chemical components which were uridine and tryptophan.The similarity results showed that the similarity of Guizhou and part of Yunnan was low,and the similarity of Hunan and Sichuan was high.The PCA analysis divided Polygonatum cyrtonema Hua into two categories,showing certain terrain and climate regularity.Hunan and Sichuan were classified as one category,Yunnan and Guizhou were classified as one category.The OPLS-DA analysis identified 3 differential markers,and ranked according to the significance of difference,which were peak 3>peak 2>peak 13.Conclusion:This method preliminarily explores the correlation between the fingerprint of Polygonatum cyrtonema Hua and different production areas,which can provide theoretical reference for the quality evaluation of Polygonatum cyrtonema Hua from different production areas.关键词
