Abstract

Objective:This study aimed to explore the effects and mechanisms of Yiyi Fuzi Baijiang Powder(YFBP)on human colorectal cancer(CRC)SW620 cells.Methods:MTT assay,scratch assay and clonogenic assay were applied to detect the effects of different concentrations of YFBP(3.5,7.0 and 14.0 mg·mL-1)on the proliferation,migration and clonality of CRC SW620 cells.RT-qPCR and Western Blot were used to detect the potency of YFBP on Hedgehog signaling pathway activity.Results:YFBP inhibited the proliferation,migration and clonogenesis of SW620 cells.Constraining Hedgehog signaling cascade,YFBP decreased the mRNA and protein expression levels of SHH,SMO,Gli1,and c-Myc but increased the mRNA and protein levels of SuFu in CRC cells.Conclusion:Overall,YFBP might suppress colorectal cancer by down-regulating Hedgehog signaling pathway activity,inhibiting the proliferation and migration of SW620 cells.
