Abstract

Objective:To investigate the effect of baicalein on angiogenesis in human trophoblast cells(HTR-8/Svneo).Methods:In this experiment,different doses of baicalein(0,5,10,20,40,and 100 nmol/L)were used to treat HTR-8/Svneo cells for 24 hours.Cell proliferation was assessed by the CCK-8 method to determine the low,medium,and high doses of baicalein.Subsequently,HTR-8/Svneo cells were treated with low,medium,and high doses of baicalein(20,40,and 100 nmol/L)for 24 hours.The supernatants of these cells were used to treat human umbilical vein endothelial cells(HUVECs)for 6 hours,and the tube formation ability of HUVECs was assessed by the matrigel tube formation assay.VEGF and PEDF levels in the supernatants of HTR-8/Svneo cells were measured using an ELISA kit.The mRNA expression levels of VEGF and PEDF in HTR-8/Svneo cells were measured by qRT-PCR.Results:Compared with the 0 nmol/L baicalein group,no significant change in cell proliferation was observed in the 5,10,and 20 nmol/L baicalein groups.However,cell proliferation activity significantly increased in the 40 nmol/L and 100 nmol/L baicalein groups(P<0.05).Compared with the 0 nmol/L baicalein group,the relative tube length and the number of cell junctions were significantly increased in the 40 nmol/L and 100 nmol/L baicalein groups(P<0.05).In comparison to the 0 nmol/L baicalein group,VEGF levels in the supernatant of HTR-8/Svneo cells were significantly increased in the 20,40,and 100 nmol/L baicalein groups(P<0.01),while PEDF levels significantly decreased(P<0.01).Furthermore,VEGF mRNA levels in HTR-8/Svneo cells were significantly increased in the 20,40,and 100 nmol/L baicalein groups(P<0.01),while PEDF mRNA levels were significantly decreased(P<0.01).Conclusion:Baicalein treatment can significantly promote angiogenesis in human trophoblast cells(HTR-8/Svneo).The underlying mechanism may involve the upregulation of VEGF levels and the inhibition of PEDF.
