基于非靶向血清代谢组学探讨参地补肾胶囊对肾间质纤维化小鼠的干预机制
Mechanism of Shendi Busen Capsule on Renal Interstitial Fibrosis in Mice Based on Non-targeted Serum Metabolomics

Abstract
Objective:To analyze the mechanism of action of Shendi Busen Capsule on serum metabolism in mice with renal interstitial fibrosis using UPLC-Q-TOF/MS and non-targeted metabolomics.Methods:Thirty 6-8 week-old male C57BL/6J mice were randomly divided into three groups:sham-operated group(Sham),unilateral ureteral obstruction group(UUO),and Shendi Busen Capsule group(SDBS).After pre-treatment for one week,unilateral ureteral obstruction surgery was performed,followed by continuous administration for 3 weeks before euthanizing the mice.Kidney tissues and serum were collected.HE and Masson staining were used to assess renal tissue pathological changes.The intervention mechanism of Shendi Busen Capsule on serum metabolic changes in UUO mice was further explored through analysis of metabolites and metabolic pathways.Results:Histopathological evaluation of kidney tissues revealed that Shendi Busen Capsule significantly improved renal morphology in UUO mice.Metabolomics analysis showed that Shendi Busen Capsule primarily intervened in renal interstitial fibrosis by modulating three pathways(glycerophospholipid metabolism,ether lipid metabolism,and the tricarboxylic acid cycle)and 14 related metabolites.Conclusion:Shendi Busen Capsule effectively improves kidney pathological changes and metabolic alterations in UUO mice,alleviates lipid disorders,and intervenes in renal interstitial fibrosis,thus reducing renal injury.关键词
