Effects of Compound Sini Decoction-Containing Serum on A549 Lung Adenocarcinoma Cell Proliferation Inhibition and Its Influence on AMP/ATP Ratio and AMPK

Abstract
Objective:To investigate the effects of Compound Sini Decoction-containing serum(XND)on the proliferation of lung adenocarcinoma A549 cells proliferation inhibition and its influence on AMP/ATP ratio and AMPK.Methods:A549 cells were divided into blank control group,AICAR positive control group,and low-,medium-,and high-dose XND groups.Except for the blank control group,all other groups were subjected to hypoxia-induced A549 cells.The XND groups were treated with DMEM medium containing 4%,8%,and 16%XND serum,respectively,while the blank control group received fresh medium and the AICAR group was treated with 300 μmol/L AICAR.Cell proliferation was measured by CCK-8 assay;cell morphology was observed under microscopy;AMP,ATP,and lactate levels were determined by ELISA;and AMPK and p-AMPK expression was detected by Western blot.Results:All XND dose groups significantly inhibited A549 cell proliferation at different time points(24,48,and 72 h)(P<0.01),with the inhibitory effect increasing with XND concentration(P<0.01).At 24 h,microscopy revealed altered morphology and reduced cell numbers in all treatment groups.At 48 h,ATP production significantly decreased(P<0.01),lactate production also reduced,with more pronounced effects in medium-and high-dose XND groups compared to low-dose(P<0.01).The AMP/ATP ratio,AMPK,and p-AMPK levels increased progressively with XND concentration.Conclusion:XND inhibits A549 cell proliferation,potentially through upregulating AMP/ATP ratio and AMPK levels to modulate cellular energy metabolism.关键词
