摘要

Abstract

Parkinson's disease(PD),as a common age-related neurodegenerative disorder,has become an urgent problem in the context of global aging,yet its pathogenesis remains incompletely understood.In recent years,growing research on genomic instability mechanisms in aging and their association with PD has provided new directions for investigating PD's pathological mechanisms and therapeutic targets.Modern TCM scholars propose that"kidney deficiency with latent toxins accumulation injuring marrow"constitutes the fundamental pathogenesis of PD,where aging-related kidney deficiency serves as the root cause.This article integrates the latest research advances to explore the correlation between genomic instability mechanisms and the occurrence/development of PD from both Western and Chinese medical perspectives,offering new insights for integrated treatment approaches.The pathological processes involving DNA damage-induced α-synuclein aggregation,neuroinflammation,and mitochondrial dysfunction belong to the"accumulation of pathogenic toxins due to kidney deficiency"process,while prolonged toxin retention leading to severe degeneration or even apoptosis of dopaminergic neurons represents the consequence of"toxin accumulation injuring marrow".The turbid toxins damage the brain marrow,deprive spiritual nourishment,impair tendon-meridian function,and cause limb dyscontrol,ultimately resulting in disease manifestation.
