从分子网络到代谢调控探究川芎和赤芍治疗缺血性脑卒中的作用机制
Mechanism of Action of Rhizoma chuanxiong and Radix Paeoniae rubra in Treatment of Ische-mic Stroke:A Study Based on Molecular Network and Metabolic Regulation

Abstract
Objective To investigate the therapeutic effect and potential mechanism of Rhizoma chuanxiong and Radix Paeoniae rubra used alone or in combination in the treatment of ischemic stroke(IS),and to compare the clinical effect of these two drugs.Methods A rat model of middle cerebral artery occlusion/reperfusion(MCAO/R)was established to simulate vascular occlusion in human IS and the process of thrombolysis and reperfusion.Network pharmacology was used to predict the potential action targets of the two drugs and related pathways,and the method of UHPLC-Q-TOF-MS/MS-based non-targeted metabo-lomics was used to investigate changes in the metabolic profile of brain tissue in rats with cerebral infarction.Results Both Rhizoma chuanxiong and Radix Paeoniae rubra improved pathological changes in MCAO/R rats,and Radix Paeoniae rubra showed a more significant effect.Network pharmacology and metabolomics analysis showed that the two drugs exerted an effect by influencing various pathways including lipid metabolism,inflammatory response,cell apoptosis,and energy metabolism.Conclusion Both Rhizoma chuanxiong and Radix Paeoniae rubra can effectively improve brain injury in IS rats,and Radix Pae-oniae rubra shows a more significant effect.The two drugs exert a neuroprotective effect through multiple targets and path-ways,and there is a synergistic effect between these two drugs,which provides new ideas and potential targets for the tradi-tional Chinese medicine treatment of IS.关键词
