基于Fe基金属有机框架纳米载药体系的构建及体外抗肿瘤活性研究
Preparation and in Vitro Antitumor Activity of Fe-based Organic Framework Nano Drug Delivery System
摘要
Abstract
Objective To construct a kind of nanomaterial with chemodynamic-chemo synergistic treatment for tumor and improve the therapeutic effect.Methods Fe-MOF@DHA nanomaterial was prepared by using metal-organic framework(MOF)as drug carrier loaded with dihydroartemisinin(DHA).The morphology and stability of Fe-MOF@DHA nanoparticles were characterized by SEM and DLS.The drug release behavior of the nanoparticle and the production of ·OH were investigated by UV-vis.The inhibitory effect of Fe-MOF@DHA nanoparticle on hepatoma HepG2 cell proliferation was investigated by MTT method.Results ·OH can be produced by the interaction of the nanoparticle with H2O2.The loading efficiency of Fe-MOF nanoparticle for DHA was 25%.The drug release rate was higher at pH 5.0.Fe-MOF@DHA nanoparticle +H2O2 group showed the best inhibitory effect on HepG2 cells.Conclusion Fe-MOF@DHA nanoparticle can be used as an ideal chemodynamic-chemo synergistic therapeutic reagent,which can effectively improve the therapeutic effect of tumor.关键词
