番木瓜离体叶片不定根发生影响因素研究
Study on Factors Influencing Adventitious Root Formation from in vitro Leaves of Carica papaya L.

Abstract
[Objective]In large scale production of tissue culture of Carica papaya L.seed-lings,low survival rate of seedlings transplanted from culture bottle caused by low rooting rate and root swelling has become the bootleneck for producing tissue culture seedlings of C.papaya.This work aimed to explore the influencing factors of adventitious root formation,establish an efficient adventitious root formation system from in vitro leaves of C.papaya,and increase the rooting rate of test-tube planlets of C.papaya,so as to provide technical support for large scale production of tissue culture of C.papaya seedlings.[Method]Using the in vitro leaves of test-tube plantlet of C.papaya as test materials,the effects of different placement methods,leaf ages and cutting methods on adventitious root formation from the in vitro leaves were studied.The contents of indoleacetic ac-id(IAA)and indole butyric acid(IBA)in different parts of the in vitro leaves were determined dur-ing the adventitious root formation.[Result]The results showed that different placement methods and leaf ages of in vitro leaves had significant effects on adventitious root formation.When the leaf upper epidermal surface(palisade tissue side)was in contact with the medium,the adventitious root formation rate was 82.2%.However,no adventitious roots were formed when the leaf lower epidermal surface(spongy tissue side)contacted the medium.The adventitious root formation rate of 30-day-old in vitro leaves was 86.2%,significantly higher than that of 60-day-old in vitro leaves,which was only 27.3%.Cutting methods of in vitro leaves had significantly differenct effects on adventitious root formation.The rooting rate of petioles alone was significantly lower than that of whole leaves.Adventitious root formed at the morphologically lower end of leaf vein inci-sions when the in vitro leaves were transversely sectioned,and the highest adventitious root forma-tion rate was observed at the base segment,followed by the middle segment,and the lowest at the top segment.Before adventitious root formation,IAA could be detected in both the top and base seg-ments of the in vitro leaves.After 20 d of induction culture,IAA was not detected in the top seg-ment,and IBA was not detected in both culture periods.[Conclusion]When the upper epidermis of in vitro leaves of C.papaya was in contact with medium,the adventitious root formation could be promoted.The ability of adventitious root formation from 30-day-old in vitro leaves was stronger than that of 60-day-old in vitro leaves.Adventitious roots formed at the base of the leaf or at the low-er end of the leaf vein incisions of leaf segments.The ability of adventitious root formation from the base segments of the leaves was stronger than that of the top segments.During the adventitious root formation in in vitro leaves,IAA accumulated at the base segments of the leaf.关键词
