Abstract

Objective:Kawasaki disease(KD)is the most common autoimmune vasculitis syndrome in children.Catalase(CAT),one of the most crucial antioxidant enzymes in the human body,plays a crucial role in maintaining free radical balance.This study aims to investigate changes in serum CAT level in children with KD and the relationship of CAT gene polymorphisms with KD susceptibility. Methods:Serum CAT concentrations were measured by enzyme-linked immunosorbent assay(ELISA)in 27 children in the acute phase of KD(acute group),27 in the convalescent phase(convalescent group),and 26 healthy children(control group).Additionally,108 KD patients and 115 healthy children were selected for genotyping of three CAT gene loci(rs1001179,rs7943316,and rs1049982)using polymerase chain reaction(PCR)and Sanger sequencing. Results:1)Serum CAT concentrations differed significantly among the acute KD group,the convalescent KD group,and the control group(all P<0.01).CAT level was significantly lower in the acute group compared to both the convalescent group and the control group(both P<0.01).At a serum CAT level of 8.55×104 pg/mL,the sensitivity and specificity for diagnosing acute KD were 63.00%and 88.50%,respectively,with an area under the receiver operating characteristic curve of 0.758,indicating high diagnostic value.2)There were no statistically significant differences in the genotype or allele frequencies of rs1001179,rs7943316,and rs1049982 between the KD and control groups(P>0.05).Similarly,no significant differences were found between KD patients with and without coronary artery lesions in terms of genotype or allele distribution(P>0.05). Conclusion:Serum CAT level decreases significantly during the acute phase of KD,suggesting CAT is involved in the pathogenesis of the disease.However,the polymorphisms at CAT gene loci rs1001179,rs7943316,and rs1049982 are not significantly associated with KD susceptibility or the presence of coronary artery lesions.
