摘要

Abstract

Objective:To investigate the possible mechanism of electroacupuncture"Sishencong"and"Fengchi"on protecting and improving hippocampal synaptic plasticity and inhibiting inflammation in rats with vascular dementia based on the NT-3(neu-rotrophic factor 3)/TrkC(tyrosine kinase receptors C)signaling pathway.Methods:The 60 SD rats after the Morris water laby-rinth test were randomly divided into the blank groups,the fake surgery groups,and the surgery groups.The four blood vessel blocking method(4-VO method)made the mouse model of the surgical group,and the 24 rats that were successfully made were randomly divided into the model group and the electronic needle group.Seven days after the molding of the electric needle group rats were taken 21 consecutive days of electro-needle intervention was done,the left and right"Sishencong"and the two-sided"Fengchi"acupoints were taken,30 minutes each time,once a day.Before the molding,after the molding,and the intervention,the Morris water maze test was used to detect the learning and cognitive ability of rats;the HE staining method was used to detect the changes in the pathological morphology of rats and hippocampus;the immunohistochemical method(IHC)detected the NL-RP3,NT-3,small gel cell IBA-1(ionic calcium connector protein)positive expression in the hippocampus;enzyme linked immu-nosorbent assay(ELISA)was applied to measure hippocampus tissue leukocyte-1β(IL-1β)and leukocytes-18(IL-18)content;Western blot was performed to determine hippocampus NT-3,TRKC,NLRP3,Caspase-1(caspase-1),SYN(synaptophysin)and PSD95(postsynaptic dense 95)protein expression.Results:The results of the water maze test showed that compared with the blank group and the fake surgery group,the model group's rats showed increased escape latency and the number of successful crossing platforms decreased(P<0.05);compared with the model group,the electronic needle group rats exhibited decreased es-cape latency and increased number of crossing(P<0.05);compared with the blank group and the fake surgery group,the IHC showed Iba-1 and NLRP3 expressions were heighted significantly(P<0.01),NT-3 significantly reduced in the model group(P<0.01);in the mice of the electrical needle group,the shape of small gel cells recovered,and the expression of Iba-1 and NLRP3 was decreased(P<0.05,P<0.01),NT-3 expression was increased compared with the model group(P<0.01);ELISA results showed that the contents of IL-1β and IL-18 in hippocampus of model group were significantly increased compared with the blank group and the fake surgery group(P<0.01);these contents were decreased in the electronic needle group(P<0.05);western blot detection showed that compared with the blank group and the fake surgery group,Heman NT-3,TRKC,PSD95,SYN pro-tein expression was significantly reduced in the model group(P<0.05,P<0.01),NLRP3 and Caspase-1 protein expression signif-icantly increased(P<0.05,P<0.01);compared with the model group NT-3,TRKC,PSD95 and SYN protein expressions were markedly upregulated(P<0.05,P<0.01)and NLRP3 and Caspase-1 protein expressions were significantly reduced in the elec-tronic needle group(P<0.05,P<0.01).Conclusion:Electroacupuncture may increase the expression of NT-3 in the hippocam-pus of vascular dementia rats,promote the binding of NT-3 to TrkC,reduce the expression of NLRP3 protein,alleviate microglia-induced neuroinflammation,improve synaptic plasticity,thus have a protective effect on the brain,and improve learning and memory ability.
