安神定志方对阿尔茨海默病认知障碍及神经炎症的影响
Effects of AnshenDingzhi Fang on cognitive impairment and neuroinflammation in Alzheimer's disease

Abstract
Objective:To observe and evaluate the effects of AnshenDingzhi Fang on cognitive impairment and neuroinflam-mation in patients with Alzheimer's disease(AD),and to verify its role in improving AD-related pathological and functional im-pairments through animal experiments.Methods:72 patients with mild to moderate AD,who met both diagnostic and inclusion criteria,were randomly categorized into two groups:the control group and the observation group.The control group was adminis-tered Donepezil therapy,while the observation group received AnshenDingzhi Fang orally in addition to the control treatment.Both groups underwent treatment for 3 months.Changes in mini-mental state examination(MMSE),AD assessment scale-cogni-tive subscale(ADAS-Cog),activities of daily living(ADL),and auditory verbal learning test(AVLT)were observed before and after treatment,and clinical efficacy was evaluated.Additionally,a rat model was prepared by lateral ventricle injection of Aβ 1-42 and subjected to corresponding drug interventions for 4 weeks.Morris and Barnes maze experiment were used to assess the learning and memory abilities of rats.Histological examination using H&E staining and Nissl staining was conducted to observe hippocampal tissue morphology and the content of Nissl bodies.Immunohistochemistry and enzyme-linked immunosorbent assay(ELISA)were employed to detect the expression of β-amyloid protein and the levels of inflammatory cytokines in hippocampal tis-sues.Results:Clinical study results showed that ADAS-Cog scores decreased significantly(P<0.05),and MMSE,ADL,and AVLT scores increased significantly(P<0.05)after treatment in both groups,with the observation group showing more pro-nounced improvements(P<0.05).Animal study results indicated that,compared to the model group,AnshenDingzhi Fang signif-icantly reduced escape latency in Morris water maze and Barnes maze,increased the number of platform crossings,time spent in the lower left quadrant,and the number of correct hole entries,all with statistically significant differences(P<0.01).Further-more,AnshenDingzhi Fang significantly increased the number of Nissl bodies in the CA1 region of the hippocampus and reduced the number of senile plaques and the levels of inflammatory cytokines,with statistically significant differences(P<0.01).Conclu-sion:AnshenDingzhi Fang exerts therapeutic effects on AD by inhibiting neuroinflammation and mitigating neuronal damage,thereby improving cognitive dysfunction associated with AD.关键词
