矾冰纳米乳调节Wnt/β-catenin通路对糖尿病型肛瘘术后创面愈合的影响
Effects of alum-ice nanoemulsion on wound healing after surgery for diabetic anal fistula by regulating Wnt/β-Catenin pathway

Abstract
Objective To observe the effects of alum-ice nanoemulsion on postoperative wound healing in diabetic rats with anal fistula and the expression of proteins related to the Wnt/β-catenin signaling pathway in rats after surgery for diabetic anal fistula,and to explore its mechanism of action in promoting wound repair.Methods Seventy-two SPF-grade female SD rats were randomly divided into sham-operated group,model group,alum-ice nanoemulsion group,and Moist Burn Ointment(MBO)group,with 18 rats in each group.A diabetic rat model was established in all groups.Subsequently,a postoperative wound model of diabetic anal fistula was created in all groups except the sham-operated group.The model group received daily local coverage with saline-soaked gauze,the nanoemulsion group received alum-ice nanoemulsion-soaked gauze,and the MBO group received MBO-soaked gauze.The treatments were applied for 21 consecutive days.The body weight,mental status,activity ability,and wound healing rate of the rats in each group were observed on the 7th,14th,and 21st days respectively.HE staining was used to examine the pathological changes and capillary density in the wound tissue.Western blot was employed to check the expression levels of Wnt1,β-catenin,Cyclin D1,C-myc,vascular endothelial growth factor(VEGF),vascular endothelial growth factor receptor 2(VEGFR2),and glycogen synthase kinase 3β(GSK-3β)proteins in wound tissue.Results Compared with the sham-operated group,rats in the model group showed decreased body weight and wound healing rate(P<0.05),increased inflammatory cell infiltration,and reduced capillary count in the wound tissue(P<0.05).On day 21,the expression levels of Wnt1,β-catenin,Cyclin D1,and C-myc proteins were significantly elevated(P<0.05,P<0.01),while the expression levels of VEGF,VEGFR2,and GSK-3β significantly decreased(P<0.01).Compared with the model group,both the alum-ice nanoemulsion group and the MBO group showed increased body weight and wound healing rate(P<0.05),reduced inflammatory cell infiltration,and increased capillary count(P<0.05).On day 21,the expression levels of Wnt1,β-catenin,Cyclin D1,C-myc,VEGF,and VEGFR2 proteins were significantly upregulated(P<0.05,P<0.01),while GSK-3β expression was significantly downregulated(P<0.01).Conclusion Alum-ice nanoemulsion may promote wound healing by activating proteins related to the Wnt/β-catenin signaling pathway,thereby improving the histopathological condition of the wound tissue.关键词
