Abstract

To address the low rate and the long time consumption of seed germination in the sexual breeding of Polygonatum sibiricum in Fujian,this study investigated the effects of different temperatures(4℃,20℃,25℃,30℃,room temperature,and variable temperature)and different concentrations(0,50,100,and 150 mg/L)of gibberellin A3(GA3)on seed germination and seedling growth of P.sibiricum.The results showed that temperature and GA3 significantly affected the seed germination and seedling growth of P.sibiricum.In the temperature test,the germination rate and germination potential reached the highest levels of 80.67%and 69.33%,respectively,after the seeds were soaked in the water at 45℃for 24 h and incubated at 25℃.Moreover,the seeds treated under this condition showed uniform germination,short germination delay,the shortest time consumption of germination,and good quality of buds.In the GA3 concentration test,when the concentration of GA3 was 50 mg/L,the germination rate and germination potential were the highest at 84.00%and 73.33%,respectively,which had no significant differences from those in the 100 mg/L GA3 group.As the concentration of GA3 increased,the seed germination delay gradually increased.The 100 mg/L GA3 group demonstrated the best growth-promoting effect,with the maximum primary bulb diameter of 3.50 mm and the most lateral roots(3.6).
