牙周膜间充质干细胞/细胞外基质复合体成骨潜能的体外研究
A comparative study of the osteogenic differentiation potential of actively and passively formed human periodontal ligament mesenchymal stem cells/extracellular matrix complexes in vitro

Abstract
Objective To evaluate the osteogenic differentiation potential of actively and passively formed human periodontal ligament mesenchymal stem cells/extracellular matrix complexes(C-MSC),and provide a theoretical basis for the active formation of C-MSC for the treatment of bone defects.Methods In vitro ex-traction of human periodontal ligament stem cells,actively and passively formed C-MSC was constructed and examined by morphological detection,hematoxylin and eosin staining;the level of apoptosis in C-MSC was detected by terminal dupt nick end labeling staining;immunofluorescence staining was used to detect the expression of osteogenesis-related proteins in extracellular matrix;quantitative polymerase chain reaction and western blotting were used to detect the expression levels of osteogenesis-related factors and proteins.Results Morphological detection showed that the actively formed C-MSC structure were denser and more elastic.Hematoxylin and eosin staining showed that actively formed C-MSC had more cells and extracellular matrices than passively formed C-MSC,and cell-cell junctions are more tightly connected.The results of ter-minal dutp nick end labeling staining showed that C-MSC were actively and passively formed,and all of them had good cell activity;immunofluorescence results showed that extracellular matrix in actively formed C-MSC was positive for osteogenesis-associated proteins.Quantitative polymerase chain reaction and western blotting results showed that the expression of alkaline phosphatase,collagen Ⅰ,Runt-related transcription factor-2,osterix in actively formed C-MSC was higher than that in passively formed C-MSC.Conclusion Actively formed C-MSC have better osteogenic differentiation ability than passively formed C-MSC,and may be more suitable for the study of bone defect regeneration.关键词
