HR-HPV PinpoRNA检测技术应用评价
Evaluation of HR-HPV PinpoRNA assay application

Abstract
Objective To evaluate whether the the high-risk(HR)human papilloma virus(HPV)PinpoRNA assay could serve as a substitute for the HR-HPV RNAscope in performing HR-HPV in situ hybridization detection on paraffin-embedded histological tissue sections by comparing the consistency between the HR-HPV PinpoRNA and HR-HPV RNAscope assays for detecting HR-HPV.Methods HR-HPV RNAscope and HR-HPV PinpoRNA were respectively used for detection in the histological sections of 68 cases,among which 47 cases had undergone HPV DNA testing by liquid-based cytology.Finally,the consistency of the two methods and that of HPV DNA and RNA detection were statistically analyzed,and the steps of the two experi-mental methods compared.Results The detection rates of HR-HPV RNAscope and HR-HPV PinpoRNA for HPV RNA in slices were 72.06%and 73.53%,respectively,and the results of the two methods were highly consistent(P<0.000 1).In terms of experimental methodology,the HR-HPV RNAscope assay requires a dedi-cated hybridization instrument and involves 6 reaction steps in its post-hybridization amplification system.In contrast,the HR-HPV PinpoRNA assay only necessitates a standard hybridization instrument with a simpli-fied 5-step post-hybridization amplification protocol.Liquid-based cytology HPV DNA testing and tissue-section HR-HPV RNA detection also demonstrated high concordance across the 47 specimens.(P<0.000 1).Conclusions The domestically developed HR-HPV PinpoRNA assay demonstrates detection efficacy highly consistent with the imported HR-HPV RNAscope kit,while offering simpler operation and lower costs,mak-ing it suitable for widespread adoption in HR-HPV RNA detection on tissue sections.Combined with liquid-based cytology HPV DNA testing,it further enhances the accuracy of pathological diagnosis.关键词
