氨甲环酸温敏水凝胶的制备及体外凝血活性评价
Preparation and evaluation of in vitro coagulation activity of tranexamic acid thermosensitive hydrogel

Abstract
Objective To prepare tranexamic acid(TXA)thermosensitive hydrogel and investigate its drug release and coagulation activity in vitro.Methods TXA thermosensitive hydrogel was prepared by cold solu-tion method using poloxamer 407(P407)and poloxamer 188(P188)as the matrix,and its preparation process optimized through orthogonal test design.The gel performance was systematically evaluated using gelling temperature,gelling time,Fourier transform infrared(FTIR)and in vitro release effect.The in vitro coagula-tion activity of TXA thermosensitive hydrogel was assessed by measuring activated partial thromboplastin time,prothrombin time,and thrombin time.Results The optimal preparation process conditions of TXA ther-mosensitive hydrogel were 20 g/mL P407+3 g/mL P188+3 g/mL TXA,its gelling temperature was about 35°C,gelling time was 166 s,the FTIR analysis confirmed the successful preparation of the P407/P188 thermosensi-tive hydrogel loaded with TXA in this study.The cumulative release rate reached 95.52%in 8 h,and the re-lease process was in accordance with the Higuchi model.The low,medium,and high-dose TXA thermosensi-tive hydrogel could all significantly shorten activated partial thromboplastin time and thrombin time,while the medium and high-dose groups could significantly shorten prothrombin time.Conclusion The TXA ther-mosensitive hydrogel exhibited excellent sustained-release effects and hemostatic functions,and has potential application prospects in controlling bleeding and promoting wound healing.关键词
