化痰活血扶正方及拆方含药血清对大鼠肝星状细胞HSC-T6增殖凋亡的影响
Effect of Medicated Serum of Huatan Huoxue Fuzheng Recipe(化痰活血扶正方)and Its Decomposed Formulae on Proliferation and Apoptosis of HSC-T6

Abstract
Objective To investigate the effects of medicated serum of Huatan Huoxue Fuzheng Recipe(化痰活血扶正方,HTHXFZ)and its decomposed formulae on proliferation and apoptosis of rat hepatic stellate cell line HSC-T6.Methods The medicated serum of HTHXFZ and its decomposed formulae,and the control group serum were prepared.HSC-T6 cells were pre-treated with medicated serum of HTHXFZ and its decomposed formulae for 24 h.The HSC-T6 cells were subdequently subjected to detect the cell viability by MTT and the cell proliferation by Edu labeling method.Annexin V/PI double staining and JC-10 staining were used to detect the effect of medicated serum on apoptosis of HSC-T6.The changes in expressions of Bel-2 and Bax were detected by Western Blot.Results MTT showed that 10％of medicated serum of the whole prescription,Huoxue Huayu Recipe(活血化瘀方)and Huatan Recipe(化痰方)had significant inhibitory effect on the growth of HSC-T6 at 24 h(P＜0.05).Edu labeling test showed that compared with the control group,the whole prescription and Huoxue Huayu Recipe sig-nificantly reduced the cell proliferation(P＜0.05).The Results of annexin V/PI showed that Huatan Recipe,Huoxue Huayu Rec-ipe and whole prescription could significantly promote the apoptosis(P＜0.01).JC-10 test Results showed that Huatan Recipe,Huoxue Huayu Recipe and whole prescription significantly increased the number of apoptotic cells in the early stage compared with the control group(P＜0.05).The expressions of Bax and Bax were decreased by Western blot(P＜0.05).Conclusion Hua-tan Huoxue Fuzheng Recipe can inhibit the proliferation of HSC-T6 and induce apoptosis through mitochondrial stress.关键词
