Abstract

Objective To examine the effect of different combinations ofHuangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma)on promoting the proliferation of the probiotic Parabacteroides distasonis(Pd),and to identify and verify the active com-ponents that enhance Pd growth through spectrum-effect relationship studies.Methods Initially,an in vitro culturing approach was utilized to investigate the impact of different proportions(3∶0,3∶1,3∶2,3∶3,2∶3,1∶3,0∶3)and concentrations(0.03125-0.5 g raw medicine/mL)of Huangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma)on the proliferation of Pd.Subse-quently,under the optimal combination concentration,a fingerprint chromatogram of ultra-high performance liquid chromatogra-phy coupled with quadrupole-electrostatic field orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap MS)for different pro-portions of Huangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma)was established.Orthogonal partial least squares re-gression analysis was applied for spectrum-effect relationship studies to screen the active components that promoted Pd prolifera-tion.The efficacy of these components in enhancing Pd proliferation was then validated.Results When the ratio of Huangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma)was 3∶1 with a concentration of 0.0625 g raw medicine/mL,the prolifera-tion of Pd was significantly enhanced,showing the strongest effect compared to the same concentration of either herb used alone.The established chromatographic fingerprint identified 23 common peaks,and 21 components were identified through standards comparison,database matching and literature references.Among these,peaks from Huangqin(Scutellariae Radix)such as baicalin(peak 11),apigenin-7-glucuronide(peak 12),chrysin 7-O-beta-D-glucopyranuronoside(peak 13),baicalin methyl es-ter(peak 14),wogonoside(peak 15),skullcapflavone Ⅱ(peak 16)and glycitein(peak 17),as well as berberine from Huanglian(Coptidis Rhizoma)(peak 9),showed a significant correlation with the proliferation of Pd.In the validation experiment of the spectrum-effect relationship,by measuring the contents of baicalin and berberine in the optimal combination(3∶1)of Huangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma),and preparing a combined solution of these concentrations,the Results confirmed the predictive findings.Furthermore,this combination was found to significantly enhance the proliferation of Pd com-pared to the same concentrations of baicalin and berberine used alone.Conclusion The spectrum-effect relationship established in this study identified the active components in the Huangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma)combination that promoted the proliferation of Pd.The active components,specifically baicalin and berberine,were confirmed through spectrum-effect validation experiments as effective in promoting Pd proliferation.This provided experimental evidence for under-standing the material basis of Huangqin(Scutellariae Radix)-Huanglian(Coptidis Rhizoma)promoted Pd proliferation,and also offered significant reference value for further development and application.
