扶正化瘀方对肝纤维化小鼠外周血及肝内淋巴细胞亚群的影响
Effects of Fuzheng Huayu Formula(扶正化瘀方)on Peripheral Blood and Hepatic Lymphocyte Subsets in Mice with Liver Fibrosis

Abstract
Objective To investigate the effects of Fuzheng Huayu Formula(扶正化瘀方)on the subgroups of peripheral blood lymphocytes(PBL)and intrahepatic lymphocytes(IHL)in mice with liver fibrosis.Methods Male C57BL/6 mice were ran-domly divided into a normal group(9 mice)and a model group(18 mice).The modeling group was fed a high-fat diet and injec-ted intraperitoneally with 10％CCl4 at a dose of 2 mL/kg,while the normal group was injected intraperitoneally with an equal a-mount of olive oil.In the third week,the modeling group was divided into a model group and Fuzheng Huayu Formula group.Dur-ing modeling,the mice in the Fuzheng Huayu Formula group were orally administered the formula(0.5 g of dry powder dissolved in 20 mL of ddH2O,at a dose of 20 mL/kg).The normal group and the model group were orally administered an equal volume of distilled water,once a day for a total of 4 weeks.At the end of the 6th week,the mice were euthanized,and serum liver function was measured.HE and Sirius Red staining were used to evaluate liver damage and fibrosis.Immunofluorescence staining of liver tissue frozen sections was performed to observe the localization of CD4+T cells,CD8+T cells and natural killer(NK)cells.Periph-eral blood and liver tissue were used to prepare single nuclear cell suspensions,and multi-color flow cytometry was used to detect the distribution of lymphocyte subgroups in peripheral blood and liver.Results Fuzheng Huayu Formula improved the liver func-tion,reduced the liver tissue inflammation and collagen deposition in mice with CCl4-induced liver fibrosis combined with a high-fat diet.Immunofluorescence staining showed that CD4+T cells,CD8+T ells and NK cells infiltrated the fibrous septa and distributed along the fibrous septa.Compared with the normal group,the model group showed a significant increase in peripheral blood CD8+T cells(0.34±0.16 vs.0.59±0.27,P＜0.05),and a significant decrease in CD4+T cells/CD8+T cells and NK/CD8+T cells(1.97±0.62 vs.1.46±0.17,4.45±1.41 vs.1.80±0.96,P＜0.05).Intrahepatic CD8+T cells in the model group were significantly increased compared to those in the normal group(3.70±1.21 vs.6.42±2.41,P＜0.05),and CD4+T cells/CD8+T cells were significantly decreased(1.05±0.14vs.0.84±0.20,P＜0.05).After treatment with Fuzheng Huayu Formula,the increase in peripheral blood CD8+T cells was less significant compared to that in the model group(0.37±0.16 vs.0.59±0.27,P＜0.05),and the decrease in peripheral blood NK cells,CD4+T cells/CD8+T cells and NK/CD8+T cells was also less sig-nificant compared to that in the model group(1.63±0.59 vs.0.96±0.30,1.73±0.36 vs.1.46±0.17,5.30±2.24 vs.1.80±0.96,P＜0.05).At the same time,the decrease in intrahepatic NK cells,CD4+T cells/CD8+T cells and NK/CD8+T cells was also less significant compared to those in the model group(4.35±0.98 vs.2.73±0.73,1.00±0.13 vs.0.84±0.20,0.73±0.21 vs.0.48±0.24,P＜0.05).Conclusion Fuzheng Huayu Formula can play a role in anti-liver fibrosis by reducing the proportion of CD8+T cells in the peripheral blood of mice with liver fibrosis,and increasing the levels of peripheral blood and intrahepatic NK cells,CD4+T cells/CD8+T cells and NK/CD8+T cells.关键词
