Exploring Mechanism of Blade Needle Therapy in Treating Muscle Injury through Autophagy Related Factors

Abstract
Objective To explore the mechanism of blade needle therapy in treating muscle injury related diseases through au-tophagy related factors.Methods Forty-eight adult SD rats were randomly divided into four groups,12 rats in each group,which were blank group,model group,acupuncture group and knife needle group,respectively.The muscle injury model was made by Bupivacaine Gastrocnemius muscle injection,and the blank group was not fed normally.The blade needle group and acupuncture group were treated with rats 2,5 and 8 days after successful modeling,respectively.After the treatment,the rats were killed with excessive anesthesia,and bilateral Gastrocnemius muscles of the rats were taken out.Autophagy related factors(Bax,TNF-α,Bel-2,LC3Ⅱ/Ⅰ,mTOR,4ebp1)were detected with HE staining,fluorescent quantitative PCR and Western blot(Wb).Results Compared with those of the blank group,the Wb detection Results showed that the expression levels of Bax and TNF-α were sig-nificantly increased in the model group,and the expression levels of Bel-2,LC3Ⅱ/Ⅰ,mTOR and 4ebp1 were decreased.Com-pared with those of the model group,the expressions of Bax and TNF-α in the blade needle group and acupuncture group were significantly reduced and the expression levels of Bel-2,LC3Ⅱ/Ⅰ,mTOR and 4ebp1 were significantly increased.The expression levels of Bel-2,LC3Ⅱ/Ⅰ,mTOR and 4ebp1 were significantly increased and the the expression levels of TNF-α and Bax were significantly decreased in the blade needle group compared to those in the acupuncture group.Compared with those of the blank group,the PCR detection Results showed that the expression levels of Bax and TNF-α were significantly increased in the model group and the expression levels of Bel-2,LC3Ⅱ/Ⅰ,mTOR and 4ebp1 were significantly decreased.Compared with those of the model group,the expression levels of Bax and TNF-α in the blade needle group and acupuncture group were significantly de-creased and the expression levels of Bel-2,LC3Ⅱ/Ⅰ,mTOR and 4ebp1 were significantly increased.The expression levels of Bel-2,LC3Ⅱ/Ⅰ,mTOR and 4ebp1 were significantly increased in the blade needle group compared to those in the acupuncture group and expression levels of TNF-α and Bax were significantly decreased.Conclusion Both blade needle and acupuncture can promote repair by regulating the autophagy function of injured rat muscle tissue,among which blade needle has a better effect.关键词
