烟草类甜蛋白TLP基因家族鉴定及表达模式分析
Identification and Expression Pattern Analysis of Tobacco-like Protein(Tlp)Gene Family

Abstract
This study systematically identified the thaumatin-like protein(TLP)gene family in Nicotiana tabacum using bioinformatics approaches and comprehensively analyzed their functional and evolutionary characteristics.A total of 11 NtTLP genes were identified from the Nicotiana tabacum genome,encoding alkaline,hydrophilic,and unstable proteins with molecular weights ranging from 42.1 to 55.4 kDa and isoelectric points between 9.12 and 9.76.Subcellular localization revealed predominant distribution in the nucleus,cytoplasm,and chloroplasts,with only NtTLP11 containing a transmembrane domain.Chromosomal distribution analysis showed that these genes are dispersed across eight chromosomes,with the highest density on chromosome Nt17(three genes).Gene structure and conserved motif analyses indicated significant intron number variation(2~8)among family members,which were classified into five evolutionary clades with distinct motif compositions.Cis-acting element analysis identified promoter regions enriched in plant hormone response,abiotic stress,and light-respon-sive elements.Collinearity analysis revealed six and 13 orthologous gene pairs between Nicotiana tabacum and Arabidopsis thaliana or potato(Solanum tuberosum),respectively,with gene duplication events driven by purifying selection.Phylogenetic analysis demonstrated close evolutionary relationships between Nicotiana tabacum TLP genes and those of A.thaliana,while tertiary structure prediction classified them into two conformational types.Expression profiling showed high expression levels of NtTLP06,NtTLP11,and others in roots,stems,and shoot apices,with NtTLP11 significantly upregulated under drought stress.This work provides critical insights into the functional characteristics and molecular mechanisms of the TLP gene family in Nicotiana tabacum resistance,offering valuable candidate genes for disease-resistant breeding.关键词
