Abstract

Wolfiporia hoelen is a commonly used bulk medicine favored by people because of its healthcare benefits and medicinal value.The market demand is great.High-quality strains are the basis for ensuring yield.However,many phenomena,such as heterogeneous germplasm resources,common homonyms,few available strains,and severe degradation,exist.There-fore,the author used 21 strains of W.hoelen from different sources as materials.By observing its growth status in the PDA culture medium and pine sawdust culture medium,the vitality status of different W.hoelen strains was analyzed.After DNA extraction,the ITS sequences were amplified via the universal primers ITS1 and ITS5 of fungal ribosome coding genes,the sequences were clustered via the maximum likelihood method with MEGA11 software,and a phylogenetic tree was con-structed to distinguish the genetic relationships.The results revealed that the mycelia of No.11,No.14,and No.20 grew vig-orously,with fast growth speeds and high overall uniformity and vitality.However,the strains of No.12 and No.13 grew weakly,their growth was slow,and the plates were not filled,indicating a tendency toward strain degradation.In terms of growth rate,the fastest-growing strain No.21,on the PDA reached 6.06 mm·d-1,and the slowest strain No.13 was only 3.60 mm·d-1;in the pine culture medium,the fastest-growing strain No.15 reached 13.25 mm·d-1,and the slowest strain No.13 only reached 6.45 mm·d-1;in terms of genetic diversity,the ITS sequence length of 21 W.hoelen strains was 834-1 090 bp,the G/C content was 58.62%-61.27%,the homology was 95.16%-99.90%,the genetic distance was 0-0.027 78,and the genetic difference was not significant.The research shows that the characteristic growth state of different W.hoelen strains is dis-tinctly different,and the growth speed of different W.hoelen strains varies between PDA and pine sawdust.Generally speaking,the strains with poor performance in PDA do not perform well on pine sawdust medium,but the strains with the best per-formance in PDA may not perform as well on sawdust medium.The ITS sequence can distinguish strains according to genetic differences,but the genetic differences among the 21 W.hoelen strains are not remarkable.The results provide a reference for screening strains planted in W.hoelen and subsequent new varieties breeding.
