4-Hydroxycinnamamide Attenuates Oleic Acid-induced Lipid Accumulation in LO2 Cells

Abstract
Nonalcoholic fatty liver disease(NAFLD)is the most common chronic liver disease in the world and is usually related to obesity and type 2 diabetes.NAFLD is a disease caused by multiple factors,and its pathogenesis is not completely clear.At present,there is no approved drug for the treatment of NAFLD,so finding safe and efficient treatments and drug development for NAFLD is crucial.Human normal hepatocytes(LO2)were induced with oleic acid(OA)to establish a NAFLD cell model,and the contents of intracellular triglyceride(TG)and total cholesterol(TC)were detected.The mRNA expression levels of the acetyl-CoA carboxylase(acc),peroxisome proliferator-activated receptor(pparγ),fatty acid synthase(fasn),stearoyl-CoA desaturase(scd-1),and sterol regulatory element binding protein-1c(srebp-1c)genes were detected via real-time fluorescence quantitative PCR.Finally,Western blotting was used to determine the protein expression levels of SREBP-1c,ACC,FASN,and PPARγ to explore the mechanism by which 4-HY improves fat accumulation in LO2 cells.The MTT results revealed that LO2 cells treated with 4-hydroxycinnamamide(4-HY)at concentrations lower than 8 µmol·L-1 had no significant cytotoxicity for 24 hours.Oil red O staining showed that OA significantly increased the formation and number of intracellular lipid droplets,indicating that the NAFLD cell model was successfully established.Compared with the blank control,4-HY effectively reduced the level of intracellular lipid deposition induced by OA in LO2 cells.4-HY decreased the levels of TG and TC in LO2 cells induced by OA,as determined by TG and TC testing kits.The real-time fluorescence quan-titative PCR results revealed that 4-HY downregulated the expression of the lipid metabolism-related genes srebp-1c,fasn,acc,scd-1,cd36,and ppar γ.Western blotting results showed that 4-HY inhibited the expression of lipid metabolism-related pro-teins such as PPARγ,SREBP-1c,FASN,and ACC.4-HY downregulates the expression of genes and proteins related to lipid metabolism in LO2 cells,inhibits the endogenous fat synthesis pathway in LO2 cells,and reduces the uptake of free fatty acids in LO2 cells,thus reducing the synthesis of TG and TC in LO2 cells and improving the lipid accumulation in the NAFLD cell model.关键词
