基于腺苷酸活化蛋白激酶信号通路探讨非酒精性脂肪性肝病发病机制及中药干预作用研究进展
Research progress on pathogenesis of NAFLD and intervention effect of traditional Chinese medicine based on AMPK signaling pathway
摘要
Abstract
Non-alcoholic fatty liver disease(NAFLD),a metabolic disorder characterized by excessive hepatic lipid deposition,is intricately linked to dysregulated energy metabolism.AMP-activated protein kinase(AMPK),a central reg-ulator of cellular energy homeostasis,plays a critical role in maintaining hepatic lipid balance.The AMPK signaling path-way ameliorates hepatic lipid accumulation and attenuates inflammatory responses in NAFLD by modulating downstream targets such as mammalian target of rapamycin(mTOR),silent information regulator 1(SIRT1),and acetyl-CoA carboxyl-ase(ACC).Recent studies have demonstrated that traditional Chinese medicine(TCM)compounds,including Shenling Baizhu powder,Erchen decotion,Gegen Qinlian decotion,and Chaihu Shugan decotion,as well as single herbs such as Zexie(Alismatis Rhizoma),Huangqi(Astragali Radix),Huzhang(Polygoni Cuspidati Rhizoma),and Renshen(Gin-seng Radix),exert therapeutic effects against NAFLD by regulating the AMPK pathway.These interventions up-regulate phosphorylated AMPK(p-AMPK)expression,inhibit lipogenic enzyme activity,and enhance antioxidant capacity,there-by modulating AMPK signaling.A systematic understanding of AMPK-associated pathways in NAFLD and the therapeutic mechanisms of TCM interventions will provide a theoretical foundation for developing multidimensional TCM-based thera-pies against this metabolic disorder.关键词
