S100A8/A9 阻滞剂ABR25757 对大鼠子宫内膜异位病灶的作用效果
Effect of S100A8/A9 blocker ABR25757 on endometriosis in rats

Abstract
Objective:To investigate the effect of the specific blocker ABR25757 on endometriotic lesions by blocking the S100A8/A9 protein in a rat model.Methods:An endome-triosis(EMT)animal model was established via intraperitoneal injection and validated.One week after modeling,Group A received intraperitoneal injections of PBS at the same frequency,with the experiment ending after three weeks.Group B started ABR25757 at 5mg/kg intraper-itoneally once daily for three days from the second week,ending after three weeks.Group C be-gan ABR25757 at 5mg/kg intraperitoneally once daily for three days from the second week,ending after four weeks.Rat weight changes,lesion volume,and microvascular density(MVD)were compared among groups.VEGF and TGF-β expression levels were detected using immuno-histochemistry and Western blot.Results:The success rate of intraperitoneal injection modeling was 90.5%.Compared to Group A,lesion volumes in Groups B and C significantly decreased(P=0.032,P=0.019).MVD in Groups B and C was significantly lower than that in Group A(P=0.000,P=0.000),and MVD in Group C was significantly lower than that in Group B(P=0.000).Immunohistochemistry and Western blot showed that VEGF expression in Group B was significantly lower than that in Group A(P<0.05),and TGF-β expression in Groups B and C was significantly reduced compared to that in Group A(P<0.05).Conclusion:Intraperitone-al injection is an effective method for modeling endometriosis.ABR25757 has a certain inhibito-ry effect on endometriosis and may represent a new treatment approach for endometriosis.关键词
