FOXM1 通过NF-κB信号通路调控宫腔粘连患者的炎症反应
FOXM1 regulates inflammatory response in the intrauterine adhesion patients through NF-κB signaling pathway

Abstract
Objective:To investigate the change of FOXM1 in endometria of intrauter-ine adhesion(IUA)patients and its role and molecular mechanism.Methods:Based on RNA-seq of endometria of IUA patients,real-time fluorescent quantitative PCR(qPCR),Western blot and immunohistochemistry of endometria were used to detect the expression and location of FOXM1 in endometria of normal controls and IUA patients.IK cells were transfected with si-FOXM1 and FOXM1 plasmid respectively,and mRNA and protein expression of inflammatory indicators(IL-1β,IL-6,TNF)were detected by qPCR and Western blot.GSEA enrichment a-nalysis were conducted in IK cells transfected with si-FOXM1.The activation of NF-κB pathway was detected by Western blot in IK cells with FOXM1 upregulation or downregulation.The ex-pression of inflammatory indictors in IK cells treated with si-FOXM1 and the NF-κB inhibitor were detected by Western blot.After induction of THP-1 cells into macrophages by PMA,the macrophages were treated with conditioned medium of IK cells(FOXM1 upregulation,FOXM1 downregulation,FOXM1 downregulation+NF-κB inhibitor),and then markers of macrophages(Arg1 and iNOS)were analyzed by Western blot.Results:The expression of FOXM1 was sig-nificantly reduced in endometria of IUA patients and mainly located in epithelial cells.Knock-down FOXM1 in IK cells upregulated the expression of inflammatory indictors while overexpres-sion of FOXM1 downregulated the expression of inflammatory indictors.After knockdown of FOXM1 in IK cells,the NF-κB pathway was activated.While overexpression of FOXM1 inhibi-ted NF-κB pathway.What's more,knockdown FOXM1 combined with NF-κB inhibitor downreg-ulated the expression of inflammatory indictors.The expression of iNOS was upregulated and Arg1 was downregulated in macrophages treated with conditioned medium of IK cells transfected with si-FOXM1.While in macrophages treated with conditioned medium of IK cells transfected with FOXM1 plasmid,they polarized to M2 phenotype.Knockdown of FOXM1 and NF-κB in-hibitor-treated IK cells reversed the pro-inflammatory polarization of macrophages induced by IK cells transfected with si-FOXM1.Conclusion:The expression of FOXM1 was decreased in en-dometrial epithelial cells of IUA patients,activating the NF-κB pathway,upregulating the ex-pression of inflammatory indicators,and promoting the polarization of macrophages towards M1 phenotype.关键词
