摘要

Abstract

[Objective]The purpose of the article was to explore the response characteristics of soil nutrient change and microbial community in tobacco planting soil with limited nutrient input in tobacco garlic rotation mode.[Method]Conventional fertilization taken as the control(NPK),five treatments including no nitrogen fertilizer(0N),no phosphorus fertilizer(0P),no potassium fertilizer(0K)and no fertilizer(0NPK)were set up to collect tobacco planting soil after flue-cured tobacco harvest and analyze soil physical and chemical properties.Based on high-throughput sequencing,the structure and function of microbial community were analyzed,and the relationship between soil physical and chemical properties and microbial community structure were further analyzed based on correlation analysis and redundancy analysis.[Re-sult]Compared with NPK treatment,the pH of 0N treatment increased by 6.0%,the organic matter(SOM)content decreased by 4.7%,and the total nitrogen(TN)content decreased by 8.6%.The pH of 0P treatment increased by 17.0%,the total phosphorus(TP)content de-creased by 19.0%,and the available phosphorus(AP)content decreased by 45.0%.The pH of 0K treatment increased by 6.0%,the total potassium(TK)content decreased by 3.9%,and the available potassium(AK)content decreased by 110.2%.The soil pH of 0NPK treat-ment increased by 13.0%,the contents of SOM,TN and TK were not significantly different,and the TP content decreased by 22.4%.The content of exchangeable calcium(Ca2+)was significantly increased under nutrient limitation.Correlation analysis showed that TP and AP were significantly negatively correlated with pH,TK and AK.The results of high-throughput sequencing showed that the diversity and richness of bacteria and fungi in nutrient-limited treatments were significantly increased,and the microbial community structure was significantly differ-ent.Correlation analysis showed that TP and AP were significantly positively correlated with Ascomycota,norank_f_norank_o_Gaiel-lales,Gemmatimonas and Neocosmospora.TP was significantly positively correlated with Actinobacteriota and Mortierellomycota.AP was nega-tively correlated with Bdellovibrionota gate.pH was significantly positively correlated with Bdellovibrionota and Basidiomycota,and signifi-cantly negatively correlated with Ascomycota,Cercophora,norank_f_norank_o_Gaiellales,Gemmatimonas and Neocosmospora.TK was significantly negatively correlated with Actinobacteriota,and AK was significantly negatively correlated with Basidiomycota,Mortierellomycota and Cercophora.Co-occurrence network analysis showed that the complexity of bacterial and fungal co-occurrence network was reduced.Nutri-ent limitation treatment increased the abundance of saprophytic nutritional function of fungi and decreased the abundance of pathologenic nu-tritional function.[Conclusion]Phosphorus is a limiting factor in tobacco-planting soil under the tobacco-garlic rotation mode.Increasing the application of phosphorus fertilizer can appropriately increase the soil pH of flue-cured tobacco stubble,promote the transformation of soil ni-trogen,potassium and Ca2+,and then regulate the soil microbial community structure,so as to achieve the purpose of maintaining a good cy-cle of nutrients in tobacco-garlic rotation farmland.
