Effects of different crop rotation combinations on bacterial community structure in tobacco-growing soil

Abstract
[Objective]The changes in the bacterial community structure and functions of tobacco-growing soil under different crop rotation combinations were explored to provide references for selecting suitable crop for compound planting patterns of high-quality and good-yield flue-cured tobacco.[Method]The Illumina NovaSeq high-throughput sequencing technology was adopted to study the impacts of different crop combinations[flue-cured tobacco-leguminous green manure → maize-leguminous green manure(D1),flue-cured tobacco-ryegrass → maize-ryegrass(D2),flue-cured tobacco-Chinese cabbage → maize-Chinese cabbage(D3),flue-cured tobacco → maize(D4,CK)]cultivation patterns on bacterial community structure and function of tobacco-growing soil during 2019-2024.[Result]The number of bacterial species in the soil under long-term rotation showed a fluctuating upward trend.The increase was significant during the maize planting season,while it was relatively small during the flue-cured tobacco planting season.The Amplicon Sequence Variants(ASVs)of soil bacteria during 2020-2024 increased by 55.9%,152.2%,108.1%,229.6% and 164.6%,respectively,compared with those in 2019.The abundance of domi-nant species increased,which were mainly Proteobacteria and Acidobacteriota.The analysis of α-diversity and β-diversity indicated that a-mong the long-term rotations of different crop combinations,D2(the ryegrass combination)had the best effect.The Chao l index increased significantly from 1735.7 in 2019 to 2362.5 in 2024.The differences in winter crops mainly affected the species at the genus level.Among them,the relative abundances of the beneficial bacterium Sphingomonas in D2 and D3 increased significantly by 10.8% and 11.79% re-spectively.After five years of cultivation,the soil microbial functions in D1 changed the least.Based on retaining nitrogen-decomposition functions such as nitrous oxide denitrification and nitrite respiration,it mainly had functions of nitrate reduction,animal parasite or symbi-ont,which was beneficial for maintaining the original functions of the soil.The changes in D2 and D3 were the most significant.D2 changed to mainly functions such as chitin decomposition,aerobic ammonia oxidation,nitrification and aromatic compound degradation,while D3 changed to mainly functions such as methylotrophy,methanol oxidation,intracellular parasitism and manganese oxidation.[Conclusion]All three crop combination patterns are capable of alleviating the suppression of bacterial diversity during the flue-cured tobacco planting season.The combination of flue-cured tobacco-ryegrass→corn-ryegrass exhibits the optimal efficacy in augmenting the variety of soil bacteria and the abundance of dominant species.The combination of flue-cured tobacco-leguminous green manure→corn-leguminous green manure is most conducive to maintaining the original microbial community and functions of the soil.关键词
